鹰爪虾加工副产物制备甲壳素关键技术研究

摘要:以鹰爪虾加工副产物为原料,通过正交试验研究有机酸结合蛋白酶法制备甲壳素工艺条件,并与酸碱法进行比较。结果表明:最佳脱蛋白条件为中性蛋白酶浓度 2 100 u/gpH 9,时间 9 h,温度 60 ℃,蛋白质去除率为79.45%±0.28%;最佳脱灰分条件为乳酸浓度25%,料液比1∶25 g/mL,时间12 h,灰分去除率为99.13%±0.57%;最佳脱色条件为过氧化氢浓度25%,料液比1∶15 g/mL,时间60 min,温度70 ℃,甲壳素白度为74.53±0.36。在此条件下甲壳素得率为11.22%±0.72%,呈白色。该法与酸碱法相比,制备的甲壳素在氢氧化钾/尿素/水体系中溶解度较高,吸湿保湿性能较好,且反应条件温和,避免了强酸强碱的使用,该研究为绿色制备技术的推广及鹰爪虾资源的开发利用提供了一定的理论参考。

Study of Key Technology on Chitin Preparation from Processing Wastes of Trachypenaeus curvirostris

AbstractThe preparation of chitin from the processed waste of Trachypenaeus curvirostris was studied by orthogonal experiment, and the results were compared with the acid-alkali method. The results showed that the optimum deproteinization conditions were: 2 100 u/g neutral protease, pH 9, 60 ℃ for 9 h, the protein removal rate was 79.45%±0.28%. The optimum conditions for ash removal were: 25% lactic acid, solid-liquid ratio 1 g:25 mL, time 12 h, the ash removal rate was 99.13%±0.57%. The optimum decoloration conditions were: 25% hydrogen peroxide, solid-liquid ratio 1 g:15 mL, 70 ℃ for 60 min, the whiteness of chitin was 74.53±0.36. The yield of chitin was 11.22%±0.72% and it was white. Compared with the acid-alkali method, the chitin prepared organic acid-enzyme methodhad high solubility in the potassium hydroxide/urea/water system, good moisture absorption and moisturizing performance, mild reaction conditions, avoiding the use of strong acids and alkalis. The research provides a theoretical reference for the popularization of green preparation technology and the development and utilization of the Trachypenaeus curvirostris.
